A challenge in H. lacustris culture is to find a way to efficiently use illumination to maintain vigorous growth and harvest dense biomass, which is essential for further exploiting the potential for astaxanthin production. Previous studies have shown that in addition to illumination, carbon supply in culture is a key limitation for algae growth. Here, we investigated a combined culture approach involving high light intensity (110 μmol m-2s-1) and injection of a 1% (v/v) CO2 air-gas mixture which provided an effective method for H. lacustris culture to achieve both a high growth rate and high cell density. The cell number in the group with high light exposure combined with CO2 enrichment was increased almost four-fold compared with a high light group (110 μmol m-2s-1 without CO2 injection). Additional experiments suggested a possible mechanism in which elevated CO2 increases the electron sink capacity, thus alleviating photoinhibition and oxidative damage. The scaled-up photobioreactor demonstrated much better performance, with growth rates improved by 50-350 %, providing further evidence that this new method can improve algal cell production. Despite this development in digital psychiatry, clinical trials are mainly based on point assessments of psychopathology. This study investigated expectable increases in statistical power by intense assessment in randomized controlled trials (RCTs). A simulation study, based on three scenarios and several empirical data sets, estimated power gains of two- or fivefold pre-post-assessment. For each condition, data sets of various effect sizes were generated, and AN(C)OVAs were applied to the sample of interest (N = 50-N = 200). Power increases ranged from 6% to 92%, with higher gains in more underpowered scenarios and with higher number of repeated assessments. ANCOVA profited from a more precise estimation of the baseline covariate, resulting in additional gains in statistical power. Fivefold pre-post EMA resulted in highest absolute statistical power and clearly outperformed traditional questionnaire assessments. For example, ANCOVA of automatized PHQ-9 questionnaire data resulted in absolute power of 55 (for N = 200 and d = 0.3). Fivefold EMA, however, resulted in power of 88.9. Non-parametric and multi-level analyses resulted in comparable outcomes. Besides providing psychological treatment, digital mental health can help optimizing sensitivity in RCT-based research. Intense assessment appears advisable whenever psychopathology needs to be assessed with high precision at pre- and post-assessment (e.g. small sample sizes, small treatment effects, or when applying optimization problems like machine learning). First empiric studies are promising, but more evidence is needed. Short-chain dehydrogenase (SDR) are a class of NAD (P) H-dependent oxidoreductases in C. testosteroni. Its main function is catalyzing the redox of the hydroxyl/ketone group of the hormone. In this paper, a SDR gene(SDRx) is cloned from C. testosteroni ATCC11996 and expressed. The polyclonal antibody was prepared and the SDRx gene knocked out by homologous recombination. Wild type and mutant C. testosteroni induced by testosterone, estradiol, estrone and estriol. The growth curves of the bacteria were measured by spectrophotometer. ELISA established the expression of SDRx protein, and high-performance liquid chromatography(HPLC) detected the contents of various hormones. The results show that the growth of wild type was faster than mutant type induced by testosterone. The concentration of SDRx is 0.318 mg/ml under testosterone induction. It has a great change in steroid hormones residue in culture medium measured by HPLC Testosterone residue in the mutant type group was 42. |
