Median distance to isocenter (DTI) was 4.7cm (range 0.2-10.5) in the SIMT group. Local recurrence-free interval was not associated with the distance to the isocenter in univariable or multivariable Cox-regression analysis. Multivariable analysis revealed only volume as an independent significant predictor for local failure (p-value <0.05). SRS/SRT using single-isocenter VMAT for multiple targets achieved high local metastases control rates irrespective of distance to the isocenter, supporting efficacy of single-isocenter stereotactic radiation therapy for multiple brain metastases.SRS/SRT using single-isocenter VMAT for multiple targets achieved high local metastases control rates irrespective of distance to the isocenter, supporting efficacy of single-isocenter stereotactic radiation therapy for multiple brain metastases. As the prognosis of early-stage breast cancer patients is excellent, prevention of radiation-induced toxicity has become crucial. Reduction of margins compensating for intrafraction motion reduces non-target dose. We assessed motion of the tumor bed throughout APBI treatment fractions and calculated CTV-PTV margins for breathing and drift. This prospective clinical trial included patients treated with APBI on a Cyberknife with fiducial tracking. Paired orthogonal kV images made throughout the entire fraction were used to extract the tumor bed position. The images used for breathing modelling were used to calculate breathing amplitudes. The margins needed to compensate for breathing and drift were calculated according to Engelsman and Van Herk respectively. Twenty-two patients, 110 fractions and 5087 image pairs were analyzed. The margins needed for breathing were 0.3-0.6mm. The margin for drift increased with time after the first imaging for positioning. For a total fraction duration up to 8min, a margin of 1.0mm is sufficient. For a fraction of 32min, 2.5mm is needed. Techniques that account for breathing motion can reduce the margin by 0.1mm. There was a systematic trend in the drift in the caudal, medial and posterior direction. To compensate for this, 0.7mm could be added to the margins. The margin needed to compensate for intrafraction motion increased with longer fraction duration due to drifting of the target. It doubled for a fraction of 24min compared to 8min. Breathing motion has a limited effect. The margin needed to compensate for intrafraction motion increased with longer fraction duration due to drifting of the target. And, in every case, there was an initial "genesis" - a "how" process that may have been at once unique to each situation and perhaps also shared a common set of facilitators. However, a full explanation of symbiogenesis also requires an answer to the "why" question, for natural selection is a stringent economizer. Something as contrarian as mutualistic cooperation between "differently named" organisms must also provide functional advantages for the participants that will be favored by natural selection (differential survival and reproduction). Enter the "Synergism Hypothesis" - the thesis that synergistic functional effects of various kinds are a common cause of cooperative relationships of all kinds in nature, including symbioses. When different organisms have complementary capabilities that are mutually beneficial and cannot otherwise be attained, the benefits derived from symbiotic cooperation will outweigh the costs. Among the many documented cases of symbiogenesis over time, lichens provide perhaps the most familiar, well-studied example, while the eukaryotes are often cited as a game-changer. The answer to the "why" question was, in each case, determinative for symbiogenesis.Recent reports have compared the analytical sensitivities of some SARS-CoV-2 RT-PCR assays, but differences in the viral materials used for these evaluations made comprehensive conclusions difficult. We carried out a direct comparison of the analytical sensitivities of 14 conventional and three rapid RT-PCR assays for the detection of SARS-CoV-2. The comparison was performed utilizing a certified reference material for SARS-CoV-2 RNA that was serially two-fold diluted in RNA storage solution. Our results show that the analytical sensitivities of the 17 assays varied within an 8-fold range (100-800 copies/mL). Moreover, a trend with some rapid assays yielding slightly higher analytical sensitivities (2- to 4-fold) compared with conventional assays was observed. We conclude that most of the RT-PCR assays can be used for routine COVID-19 diagnosis, but some assays with the poorest analytical sensitivities may lead to false-negative results when used to identify asymptomatic individuals who can carry a low viral load but still be infectious. These findings should be kept in mind when selecting high-sensitivity and rapid assays. The World Health Organization (WHO) has declared a pandemic of COVID-19, the disease caused by the recently described SARS-CoV-2. The relevance and importance of mass diagnosis in order to find the asymptomatic individuals is widely recognized as a mandatory tool to reinforce the control measures for monitoring virus circulation and reduce the spreading of SARS-CoV-2. https://www.selleckchem.com/products/pt2977.html Here, we described quickness and cheaper strategies of direct RT-qPCR (in the absence of RNA isolation) and compared the results to those obtained using standard RNA isolation procedure. The tests varied using pure, diluted samples, combined with Proteinase K (PK) or Lysis Buffer. Our findings showed consistently that PK pre-treated samples in the absence of RNA extraction procedures presents similar results to those obtained by standard RNA isolation procedures. On average, 16 samples extracted with the MagMAX™ CORE Kit, take around 2 h, costing an average of USD 5, the pre-treatment of samples using PK, on the other hand, would cut the value to less than USD 0.30 and reduce the time of procedure in more than 1 ½ hours. The present study suggests the use of PK treatment instead of RNA isolation in order to reduce costs and time in processing samples for molecular diagnosis of SARS-CoV-2.
